Abstract

Ferrokinetic studies in man have been undertaken by liquid scintillation counting of 59Fe in plasma and whole blood. Measurements of radioiron were also made at the body surface, on the sacral bone and over the liver and the spleen. A seven compartment model was formulated and used in analysing the experimental data. After assessing the structural identifiability of the model, its parameters have been identified by a gradient method. Moreover, the reliability of the obtained estimates has been evaluated. The usefulness of the more precise data collection procedure based on liquid scintillation counting rather than on the conventional method for 59Fe gamma counting has been appreciated. Data from 7 normal subjects are here analysed in terms of the proposed model. For each of them, the obtained parameter estimates and the corresponding calculated pool sizes and transfer rates between compartments are presented and discussed. The model proved adequate for discriminating some anemic states on the basis of the changes of its identified parameters; the results obtained from a limited number of patients affected by aplastic and dyserythropoietic anemias are also reported.
