Anticonvulsant activity and brain levels of diazepam and its metabolites in mice

Abstract

The i.p. LD5 0 and the i.v. ED5 0 against pentetrazol were determined in mice for diazepam, N-demethyldiazepam, N-methyloxazepam and oxazepam. After administration of the ED5 0 the brain concentrations of these benzodiazepines were related to the brain levels of the injected drug only at 5 min. At later times the effect was related to the brain levels of the metabolites; this was particularly true of oxazepam, the benzodiazepine with the best therapeutic index.
