With the aid of serial microphotographs and phase contrast microcinematography, studies were carried out on cultures of histiocytes and macrophages of Molgevulgaris to determine the morphological and functional relationships existing in the interphase between the nucleus and the centrosome. Two functional aspects of the centrosome are described: the coordination of movements of the granulations within the cytoplasm and in the cell as a whole, and the maintenance of close connections between the centriole and the nuclear structures. These latter may be lost under certain functional conditions, while the coordinating function remains unaffected. The results are discussed in the light of certain hypotheses on the persistence during the interphase of connections between the centrosome and the nucleus.
