Different neuroectodermal cell lines differentiated by retinoic acid express various protein isoforms of the amyloid precursor protein (APP). Human neuroblastoma [SK-N-BE(2), SK-N-SH, SH-SY5Y, CHP 100, SMS-KNR], and melanoma [SK-Mel 28, TVM-BO] cell lines produced two major APP of about 103 and 93 kDa. The glioblastoma line [LI] expresses only the larger APP isoform. With one exception, all cell lines showed only minor augmentation in intensity and ratio between the two isoforms after 5 days exposure to 5 μM retinoic acid. However, SH-SY5Y cells showed the induction of a smaller APP isoform of about 81 kDa accompanied by a relative reduction in intensity of the two larger proteins. These data were also confirmed by immunohistochemistry, which also showed no changes in intracellular localisation of APP, but with considerable cell-to-cell heterogeneity suggesting heterogeneous cell regulation.
