High sensitivity optical microscppe for single molecule spectroscopy studies

Abstract

The single molecule emission with 50 ms time resolution was investigated with a closed loop piezo actuator and detection was performed through avalanche photodiodes. The two algorithms was used for fast imaging in raster scanning two photon microscope. A fluorescence fluctution analysis was implemented in slow scanning algorithm by computing the photon counting histogram (PCH) on each pixel of the image. The point spread function of the setup was measured and applications to cell imaging and single dyes were described.
