Improvements in megakaryocyte purification

Abstract

An improved two step procedure to purify megakaryocytes (MK) is proposed. Guinea Pig bone marrow particles are incubated with collagenase to obtain a single cell suspension which is further processed by buoyant density gradient centrifugation and by velocity sedimentation using a modified sedimentation chamber. An high degree of MK purification is achieved. Purified MK were investigated by Scanning Electron Microscopy (SEM). Our observations provide new insight on the surface structure of these cells.
