Lack of presence of Human Papillomavirus DNA in amniotic fluids from pregnant women with latent genital infection

Abstract

Objective: To examine the possibility of intrauterine transmission of human Papillomavirus DNA from mothers with genital latent infection. Methods: Ninety-three pregnant women between 15-16 weeks gestation were tested following their examination in the obstetric unit for genetic reasons. Polymerase chain reaction was used to seek human Papillomavirus in maternal cervico-vaginal lavages and amniotic fluid samples. Amplified products of amniotic fluid samples from mothers with genital latent human Papillomavirus infection were further investigated by Southern Blot. At birth, the infants born to mothers with genital latent Human Papillomavirus infection were further investigated for the presence of HPV-DNA in the nasopharingeal aspirates. Results: None of the 93 amniotic fluid samples tested positive for human Papillomavirus DNA either after polymerase chain reaction amplification or after Southern Blotting. In mothers human Papillomavirus infection was identified by consensus primers in 8 (8.6) cases. By type-specific primers HPV 16 was detected in 2 women tone of them also positive for human Papillomaviruses 6 and 11), human Papillomaviruses 6 and 11 were detected in 4, and human Papillomavirus 33 in two. At birth the oral samples from the 8 infants born to human Papillomavirus positive mothers were negative. Conclusions: Our results suggest that transplacental transmission of human Papillomavirus infection from mothers with latent infection is an unlikely event.
