Microphotometric and morphometric evaluation by image analysis of human Y79 retinoblastoma cells over time in culture

Abstract

A microphotometric and morphometric study by image analysis on human Y79 retinoblastoma cell nuclei was carried out at 24 hour intervals over a ten day period of in vitro culture with no change of the medium. For every twenty-four hour time period, two different sets of 500 Feulgen-Thionine stained nuclei were measured on the same slide. Overall, 10.000 nuclei were measured. Evidence of diploid DNA content in 9 of 10 culture times was confirmed by the modal diploid chromosome number using cytogenetic analysis. The microphotometric analysis of the DNA content showed a consistent hypodiploid cell population which was prevalent in 2 of 10 culture time periods. The average optical density was high and showed an increase with time in culture. Nuclear size varied significantly, while nuclear shape varied only slightly and both were rather similar in later culture times. Scatterplots of DNA mass distribution versus nuclear size demonstrated that the Y79 retinoblastoma cells have a heterogeneous nuclear structure. Varying nuclear size and structure were detected in diploid and tetraploid cells and during the transition from diploid to tetraploid cells. In conclusion, the combination of microphotometric and morphometric analysis permits morphologic heterogeneity of human retinoblastoma Y79 cell line to be demonstrated objectively.
