Morphine concentrations in the rat after intraperitoneal or subcutaneous injection

Abstract

Morphine sulfate was administered to adult male albino rats in a dose of 4 or 64 mg/kg by ip or sc injection. Plasma and brain levels of free morphine were measured at times up to 3 hr post-injection. Total morphine levels were measured up to 1 hr after injection. Peak brain levels of free morphine after 4-mg/kg injections occurred at 30 min and were 4 times higher for sc than for ip; total morphine concentrations at that time were not statistically different. After a 64-mg/kg dose, free morphine levels in plasma and brain peaked at 1 hr, the ip levels being somewhat higher than the sc. Total morphine concentrations in plasma were not significantly different for 64-mg/kg doses at 1 hr. The data presented show that animals given equal mg/kg doses of morphine may be exposed to different amounts of pharmacologically active drug depending on the route of administration.
