A radioactivity dilution technique is described which permits easier and more precise determination of the ascitic fluid volume, and hence of total cell numbers, than the usual method employed. Since within 20 min after ip injection no significant radioactivity was detected in the portal and peripheral blood, indicating no substantial leakage of label from the peritoneal cavity, the assay could also be applied in living animals. It has recently been used with success in chemotherapy and tumor immunology studies.
