Promoter polymorphisms of the 5-HT2C receptor gene

Association study with panic disorder in two independent samples

Abstract

The 5HT2C receptor gene has been implicated in the pathogenesis of panic disorder because the nonselective 5HT2C receptor agonist m-chlorophenylpiperazine (mCPP) causes panic attacks in challenge tests. Two novel polymorphisms have recently been described in the 5′-region of the 5HT2C receptor gene, a (GTU2-18 and a (CT)4-5 polymorphism separated by a 54 bp spacer (Meyer et al., Med Genet 11:200, 1999). The constitute one short (88bp) and several long allele haplotypes (94-100bp). In a first association analysis in a German sample (n=79), genotypes containing only long alleles were significantly more frequent among women with panic disorder than among women of a control population (p=0.04, Chi Square analysis). A second analysis in an independent Italian sample (n=124), however, did not show any significant difference in the number of genotypes with only long alleles between women with panic disorder and women of a control population (p=0.54). Probably, the finding in the German sample is a false positive. This is consistent with the failure so far to detect any functional difference between the different allele haplotypes. Other explanations for the nonreplication are a small size of the effect or differential linkage disequilibrium to a nearby true functional polymorphism in different populations.
