Study of correlation between a microplate system and a traditional one to evaluate ALT as parameter for the validation of the blood unit

Abstract

For the validation of blood unit five tests are requested for Italian law: HBsAg, anti HTV 1-2, ami HCV, Syphilis and ALT. With this regard, it was introduced a system to manage automatically the five tests, which includes a dedicated form for the determination of ALT (microplate method). The main purpose of this study is to evaluate the affidability of this new test comparing it with a traditional one (method IFCC 37°C). 1315 Donors' samples were analyzed with the following different methods: • method IFCC (37° C BMI) using HITACHI 704 instrument • kinetic enzymatic fixed time in microplate method (Ortho ALT microwelll test) using ORTHO SUMMIT instrument, including the following forms : Genesis Tecan, Autoreader in and software Medusa 2000. The results demonstrate a good correlation between the two systems used (r=0,959491) and a very good reproducibility [The results in the fig. 1 show values of CV referred to the two control sera with enzymatic activity for Alt about 53 U/l 37°C (CV=3,5%) and 70 U/l 37°C (CV=3,8%)]. The use of Ortho ALT Microwell test on Ortho Summit would permit to all Blood Banks to centralize and make in a very fest way the five tests necessary for the validation of the Wood unit.
