Three hundred and sixty one courses of cisplatinum were given in a one-hour regimen of hydration with mannitol-furosemide-induced diuresis to 86 patients with advanced malignant tumors. Doses were 75 mg/m2 iv every 3 weeks as a single agent in 80 patients and 60 mg/m2 iv in combination with adriamycin and VP-116-213 in 6 patients. Reversible nephrotoxicity occurred after 16 courses (4,43%) in 9 patients (10,46%). No patient developed an increase of BUN > 60 mg/dl or serum creatinine > 3.0 mg/dl. Cumulative renal damage, loss of hearing, and peripheral neuropathy were not observed. Nearly all patients experienced nausea and vomiting. Myelosuppression was mild.
